Learning outcomes

Life in all its forms is based on nucleic acids which store and transfer genetic information and catalyze some fundamental cellular processes, including protein synthesis and tailoring of nucleic acids. Accordingly, nucleic acids have received increasing interest as a target of chemotherapy and oligonucleotides as chemotherapeutic agents. This lecture course is aimed at elucidating the central role of chemistry in the rapid developing field of nucleic acid cell biology. Attention is paid to synthesis of structurally modified biomolecules and their conjugates useful as research tool for cell biology and understanding of the chemical basis of biological phenomena at the level of individual bond making and breaking processes and interatomic interactions.


Protolytic equilibria, conformations, tautomerism and coordination chemistry of nucleosides and nucleotides. Structure and reactivity of nucleic acids. Basic principles of phosphorus chemistry. Catalytic nucleic acids. Synthesis of nucleosides, nucleotides and oligonucleotides, their structural analogs and conjugates useful in chemotherapy or research tools for cell biology. Therapeutic oligonucleotides. Pro-drug strategies of nucleotide drugs.
