Learning outcomes

The course teaches students how to use UV, MS, MS/MS, CD and multiple NMR techniques to fully unravel structures of bioactive compounds. At the same time students learn how to determine at least three bioactivity types for the unravelled structure by several bioactivity methods.

Contents

The course starts by collecting a large mass of bioactive plant material that is left to macerate in a suitable solvent. After maceration, the extract is freeze-dried and subjected to preparative and semi-preparative HPLC purification steps. Purification is followed by UPLC-DAD. Pure compound is freeze-dried and analysed for UV, MS & MS/MS (Orbitrap), CD, and numerous 1D and 2D NMR spectra. The obtained spectra are inspected in detail to characterize the full structure. The bioactivities will be measured by seven different methods for protein affinity, oxidative activity and antioxidant activity.
