Learning outcomes

The course aims at teaching the student how different compounds are purified by preparative and semi-preparative HPLC both in theory and practise. The student also learns how to optimize and create a quantitative UPLC-MS/MS method for any compound that is ionizable via electrospray ionisation. At the same time the student learns to understand how MS/MS technology can be used to benefit structure characterization of unknown compounds.

General description

The course deals with methods for large-scale (>100 mg) isolation and purification of different compounds and uses preparative and semi-preparative HPLC methods also in practise. Purified compounds are used to create and optimize a quantitative UPLC-DAD-MS/MS method. The developed method will be used to find limits of detection, limits of quantification and linear range for quantification for various analytes by (1) diode array, (2) full scan, (3) SIM (selected ion monitoring) as well as (4) MRM (multiple reaction monitoring) quantitation techniques.
